Madsen struck 111, while Matt Critchley (85) and Billy Godleman (52) also made half-centuries on a good day for the visitors at The 1st Central County Ground, who closed on 371-5.
Brooke Guest (30 not out) and Anuj Dal (five not out) will resume on Day Two, with 14 overs to secure a fifth batting point.
